This study examined the effects of dietary glycoprotein extracted from Porphyra yezoensi on growth performance and resistance against the pathogenic bacteria Edwardsiella tarda in olive f lounder. A porphyra-originated glyco- protein (P) was extracted using sequential processes of water and ethanol treatment. P extracts were added to a f ish- meal-based diet at concentrations of 0.0, 0.5, and 1.0% (designated as Con, P0.5, and P1.0, respectively). Fish were fed one of the three experimental diets for 10 weeks. All f ish groups exhibited over 96.7% survival during the ex- perimental period. Results indicated that the f ish fed diets containing P showed an increase in growth performance, including enhanced weight gain, specif ic growth rate, and feed eff iciency. An increase in insulin-like growth factor (IGF-1) was observed in the f ish fed the P1.0 diet, as compared to those fed Con. At the end of the 10-week feeding trial, all f ish were infected with E. tarda, and accumulated mortality was monitored for 8 days. Fish fed the Con diet exhibited increasing mortality from day 3 to the end of the challenge test, whereas the mortality of P-fed f ish ceased at day 5. We suggest that supplementation with P-originated glycoprotein in aquafeed may increase growth performance and resistance against pathogenic bacteria in olive f lounder juveniles.

서 론

최근에는어류의성장증진목적이외에사료효율, 면역능력 어체일반조성향상을목적으로배합사료에여러기능성 질을첨가한배합사료개발연구가활발히이루어지고있다 (Kim et al., 2011). 특히, 단백질합성과세포성장을촉진시킬 있는사료첨가물연구는배합사료를이용한양식어류의 장을극대화시키는측면에서앞으로지속적으로수행될연구

주제일것이다.

사료첨가제를이용한배합사료개발에는양식어의성장 진은물론면역력증강을통해양식어의패사율을낮추는목적 으로도연구되고있다. 최근기능성사료개발을위해천연물 에서기능성물질을찾고자하는연구가활발히진행되고있다 (Kim et al., 2006; Cho et al., 2006). 따라서연구에서도 연물중하나인우리나라해조류중에서생산량이높으며단백 함유량이높은김을이용하여새로운기능성배합사료소재

개발하기위해시도하였다. 김은홍조식물보라털목보라털 과의해조류로서한국, 일본, 중국의연안에분포하고양식도 가능하여쉽게접할있는해조류중의하나로서최근해조류 관한관심이높아지면서이에따른연구도활발히진행되는 중이다.

해조류는식품으로서뿐만아니라그들이가진다당류의 능성으로인해많은연구가진행되고있으며, 특히해조류

김과같은홍조류는 10-47% (건조중량)단백질을함유하고

있어식물에서는중요한단백질원으로보고되고있다(Fleur- ence, 1999). 홍조류인 Porphyra umbilicalis 에서추출한

당류인 porphyran주로아시아에서소비되고있으며, 콜레

스테롤수준을감소시키고항생제와항암효과를가지고있으 (Koo et al.,1995; Hong et al., 1997), fucoidan 항염증 항산화효과를가지는것으로보고되고있다(Nishino et al., 1991; Takashi and Terukawu, 1991; Koo et al., 1997). 한편, 방사무늬김Porphyra yezoensis 추출물인당단백질은 rat 대상으로 in vivo 상에서손상에대한회복기능(Hwang et al., 2008)항염증효과를가지는것으로보고되어(Shin et

al., 2011) 해조류가가진생리활성물질에대한연구가활발히

진행되고있는것을있다. 최근에는이러한해조류가 생리활성물질을어류양식에적용시키고자하는연구가 행되고있으며, Mustafa et al. (1995) red sea bream대상 으로유효성을연구한있다.

현재까지동물 세포의성장인자는많이 알려져있으나 단백질영양동물성장과밀접한관련이있는 Insulin-like growth factor-I (IGF-I) 70개의아미노산으로이루어진 polypeptides로서 proinsulin구조적으로유사하다 (Spark- man et al., 2012). IGF-I척추동물에서성장호르몬의매개로 생성되어동물의성장에중요한역할을하고있으며세포의 , 분화, 대사에다양한효과를보여주고있다 (Baxter, 1993).

Insulin유사한대사효과를발휘하면서도 (Caro et al., 1988) insulin과는달리 IGFs주로간에서생성, 분비되면서 뇌하수체, 난소, 등과같은다른기관에서도생성되며체성 장을가속화시키고여러조직의성장을선택적으로유도할

아니라in vivo에서단백질의합성을촉진시킨다.

이전실험에서는김에서추출한당단백질을세포를이용한 실험에서정상근육세포의성장을촉진시킨결과를확인하였 세포수준에서확인된성장효과가 in vivo 상태에서넙치의 성장과면역에도영향을미칠것이라는가정하에실험을진행 하였다.

재료 및 방법

김으로부터 당단백질의 추출

2010 5월에전라남도완도군에서채취하여말린건조김을 사용하였고당단백질추출과정은 Fig. 1나타내었다. 분쇄된

분말 600 g대용량추출용기에넣고증류수 15 L침지 하였다. 실온에서 3시간동안교반하면서추출한다음, 5,000

rpm 4℃에서 20분간원심분리하여상층액을취해 3부피의

에탄올을첨가하고침전된당을뷰흐너깔대기로제거한 액을취해감압농축하였다. 완전히농축하여에탄올과수분을 제거한시료에다시증류수를첨가하여용해시킨다음이를 결건조하여실험에사용하였다.

실험어 및 사육조건

실험에사용된실험어류는경남통영스마일사료거제양식장

에서수송된넙치육성어를운반하여 10 ton 원형수조에서

환경에적응할있도록 2주간예비사육을실시하였으며, 예비사육기간동안상품배합사료를공급하였다. 예비사육 넙치육성어 (초기평균중량: 92.5±2.21 g) 18개의 2 ton PP원형수조에수조 30 마리씩무작위로배치하였고 료공급실험은실험구당 3 반복구를두었으며, 사육수는여과

해수를사용하여 3-5 L/min 유수량이공급되도록조절하였

. 모든실험수조에용존산소유지와원활한사육수순환을 위하여에어스톤을설치하였다. 실험기간동안평균수온은 21.8℃ (18.5-24.1℃) 범위로자연수온에의존하였으며, 실험 사료는 1 2(09:00 h 16:00 h)나눠서 10동안만복 공급을실시하였다.

실험사료

넙치실험에사용된실험사료의사료원료일반성분조성 Table 1나타내었다. 실험사료는(P. yezoensis) 당단백 (glycoprotein) 0.5% (P0.5), 1% (P1.0) 농도별로첨가한 실험사료와당단백질을첨가하지않은대조사료(Con) 계하였다. 실험사료의원료로는어분, 탈피대두박을주요 백질원으로이용하였고, 소맥분을주요탄수화물원으로이용 하였으며, 오징어간유를주요지질원으로사용하였다. 대조구 제외한당단백질사료에는각각의첨가제원료를셀룰로 대신에첨가하였다. 실험사료제조는우선모든사료원들을 파쇄기를이용하여분말형태로일정하게만들고, 사료원들 사료조성표에따라정확히중량을, 섞은다음사료

총량의 30-40%해당하는증류수를첨가하여사료혼합기

(NVM-14-2P, KOREA)혼합, 반죽시켰다. 혼합반죽물은 형쵸파기(SMC-12, KOREA)이용하여직경 3 mm 크기로 성형되었다. 성형된실험사료는 -70℃ 동결냉동건조기에서

건조시켜, 시브(sieve)이용하여적당한크기의사료로가공

되었으며, 사료공급전까지 -20℃ 냉동고에보관되었다. 성장도 조사

어체무게측정은 5간격으로실시하였으며, 24시간절식

시킨 MS-222 (100 ppm)마취시켜전중량을측정하였다. 실험종료후, 증중율(weight gain, WG), 사료효율(feed effi- ciency, FE), 일간성장률(specific growth rate, SGR) 생존율 (survival rate)조사하였다. 일반성분은실험사료와전어체 AOAC (2000)방법에따라수분은상압가열건조법(105℃, 4시간), 조단백질은 Kjeldahl 질소정량법(N×6.25), 조회분은 직접회화법으로분석하였다. 조지방은샘플을 12시간동결 조한, Soxtec system 1046 (Tacator AB, Sweden)사용하 soxhlet 추출법으로분석하였다.

혈중 IGF-1 활성

혈액중의 IGF-1 activity측정하기위해 Human IGF-1 EIA kit (K032112, Komabiotech, Korea)사용하여분광광 도계(Ultraspec 2001 pro. Amersham Phamacia biotech, Eng- land) 450 nm에서흡광도측정하였다. , Antigen-affinity purified Goat anti-Human IGF-I pre-coated 96-well plate standard혈청을 100 μL 첨가하여실온에서 2시간반응시킨 PBS세척하였다. 0.32 μg/mL detection antibody well

100 μL첨가한실온에서 2시간반응시켜세척한후에

color development enzyme (1:20 dilute) well 100 μL 가하여실온에서 30반응시켰다. 반응이완료되면세척하고, color development solution 100 μL 첨가하고실온에서색이 변하면 stop solution첨가한다음 micro plate reader (Bio-

Rad, USA)사용하여 450 nm에서흡광도를측정하여대조

군을백분율로환산하였다. 병원성 균주 저항성 실험

당단백질첨가제를공급한넙치의병원성세균에대한 항성을조사하기위하여 Edwardsiella tarda 사용하여공격실 험을수행하였다. 실험구별로 5마리씩에대하여 E. tarda GY-01 strain (0.99×106 - 2.8×106 CFU/fish)복강내주사 하여대조구에대한상대생존율조사하였다.

통계처리

실험의분석결과는각각의군별로평균과표준편차(mean

± S.D.)사용하여표기하였으며, 모든자료는 Windows SPSS 프로그램(Statistical Package for Social Science, SPSS Inc, Chicago, IL, USA)이용하여처리하였고, 반복측정에 의한 ANOVA test Duncan's multiple range test 의성검증을실시하였다. 모든통계적유의도수준은 P < 0.05 에서결정되었다.

당단백질을사료첨가제로서사용하여넙치의 10주간 체율, 사료효율, 일간성장률, 일간섭취율, 생존율의실험결 성분분석결과는 Table 2 - 4나타내었다. 일간사료섭 취율생존율에있어서는모든실험구에서유의한차이를 타내지않았다(P>0.05). 당단백질실험구는대조구와비교 하여어체중의유의적인증가가나타났으며, 당단백질 험구(P0.5 P1.0)대조구(Con)비해각각 10%, 11%

가되었다(P>0.05). 일간성장률와사료효율에서도당단백

실험구(P0.5 P1.0)대조구(Con) 보다유의하게높은경향 나타내었다 (P>0.05). 따라서, 당단백질의섭취가넙치 성장에영향을미치는것을있었다. 이와유사한결과가 yellowtail대상으로한실험에서보고되었다. Fukada et al.

(2009) 40일간의P. yezoensis 첨가실험을수행한결과사료 2% 첨가실험구에서성장과사료효율이개선되는경향을 보고하였다. 한편, 넙치에서감태를사료내첨가하였을때는 성장이감소하였으며(Kim and Lee, 2008), 무지개송어에서도

P. dioica사료내첨가하였을성장이감소하는결과를

였다(Soler-Vila et al., 2009). 이와유사한결과는 grey mullet (Davies et al., 1997), red sea bream (Mustafa et al., 1995), European sea bass (Valente et al., 2006)에서도보고되었다. 나일틸라피아에서는 P. yezoensis농도에따라서성장이 감하는것을보고하여(Stadtlander et al., 2012), 해조류의종류 농도어종에따라서도영향이다르게나타났다. 그러 연구에서는당단백질이효과적인것으로나타났으며, 실험기간이 10주인것을고려해보았을장기간당단 백질을첨가한사료를공급한다면더욱확연한차이를나타낼 것으로생각되며출하시기역시앞당길있을것으로기대된 .

혈액중 IGF-1 활성

Insulin-like growth factor type 1 (IGF-1)성장촉진과관련 생화학적인지표로서 IGF-1 70개의아미노산으로구성 되어있는 single chain polypeptide증식세포분화촉진 다양한역할에직접적으로관여하는성장인자이다(Spark- man et al., 2012). 간에서생성되며혈액을통해전신으로운반 되어근조직의형성에도중요한역할을하는것으로밝혀져 으며, 최근연구에서는 Chilean flounder (Paralichthys adsper- sus)에서 IGF-1 IGF-1 Recepter유생발달신경시스템, 근육, 구조형성에도중요한역할을하는것으로보고되었 (Escobar et al., 2011).

이전에보고된연구에서는넙치의크기를달리하여혈액 IGF-I함량을측정하였을체중의증가에따라 IGF-I 량이달라지는결과가보였다(Nam et al., 1996). 연구에서 사육한넙치혈액을채취하여서 IGF-1측정한결과(Fig.

군에서농도유의적으로 IGF-1분비와활성이높은 으로관찰되었다. , 당단백질의첨가로인해혈중 IGF-I 활성이높아졌으며, 이는당단백질을섭취한넙치가대조구

비해10-11% 어체중증가의결과를나타낸것으로사료

된다. 따라서, 당단백질을섭취한넙치가일반사료를섭취 넙치보다성장속도가빠를아니라면역증강의효과 가져올것으로예상된다. Fukada et al. (2009) 40일간의 Porphyra yezoensis 첨가실험을수행한결과사료내 2% 첨가 실험구에서성장과사료효율이개선되는경향을보인바있다. 간에서 Fructose-1,6-bisphosphate, phosphogluconate dehy- drogenase, glucose-6-phosphate dehydrogenase, 흑색근에서 carnitine palmitoyltransferase II 효소활성이함께증가한 으로보고되었다. 이와같은효소활성은 IGF-1영향과직접 적으로관여되며, 실험에서의성장개선효과와관련된것으 사료된다.

병원성 균주 저항성 실험

E. tarda그람음성의장내세균과의주요어병세균으로

류에 edwardsiellosis유발하는전신감염성병원체로서

식산업에커다란피해를주고있다(Miwa and Mano, 2000).

E. tarda균은넙치에감염되어질병을일으키는데(Bang et al.,

1992), 일반적인임상증상으로서복수에의한 복부팽만,

구백탁, 체색흑화, 탈장, 항문부위의출혈, 그리고, 신장, 비장이나아가미에작은흰색결절형성등으로알려져있다 (Mohanty and Sahoo, 2007). 10동안당단백질을함유한 사료를급이하고질병이없는건강한넙치 10마리씩을무작위

추출하여 E. tarda 세균에감염시킨누적폐사율변화를

Fig. 3나타내었다. 해조추출물인당단백질첨가에따라

어병세균에대한저항성을알아보기위해실시한공격실험 , 접종한지 3일째급격히폐사하는경향을보였으나, 5일째 부터는당단백질실험구들에서는대부분폐사하지않았다 (Fig. 4). Kim et al. (2002) Choi et al. (2004) aloe 0.5% song-gang stone 0.5% 사용한결과 E. tarda대한내병성이 있는것으로보고하였다. 이와같은결과는실험에서사용한 사료당단백질의첨가가넙치의성장을촉진시킬뿐만 아니라김에포함된생리활성물질이넙치의특이적면역반

응을증강시켜 E. tarda질병에대한저항성을향상시키는

으로사료된다. 따라서, 당단백질의사료첨가는넙치의 성장내병성에좋은효과를나타낼것으로생각된다.
