Background: Increased oxidative stress play an important role in the development and perpetuation of the asthmatic airway infl ammation. A potent methyl donor, S-adeno- sylmethionine (SAMe) is known to have protective function in liver injury and fi brosis through modulation of oxidative stress. The aim of this study is to evaluate the effect of SAMe on airway infl ammation in a murine asthmatic model.

Methods: Asthmatic mice were generated by intranasal (i.n.) administration of oval- bumin (OVA) with fungal protease twice a week for 8 weeks and were challenged with aerosolized OVA. The asthmatic mice were divided into 3 groups and orally treated with vehicle, low dose SAMe (30mg/kg/day per mice), and high dose SAMe (150mg/kg/

day per mice) for each group before each OVA challenge. Bronchoalvelolar lavage (BAL) fluid analysis and histopathologic examination were performed to evaluate airway infl ammation. Various cytokine were quantifi ed in lung tissue. Oxidized glutathione (GSSG) and markers of lipid peroxidation in lung tissue were also measured.

Results: Infl ammatory cells in BAL fl uid, tissue infl ammation and fi brosis were reduced in both treatment groups and especially effective in high dose group. Th2 pro-infl am- matory cytokines such as IL-5 and IL-13 also decreased in treatment groups. The levels of GSSG and markers of lipid peroxidation were reduced in treatment groups.

Conclusions: These results demonstrated the suppressive effect of SAMe on airway infl ammation and fi brosis in mouse model of asthma and suggest its potential as new therapeutic agent in asthma treatment.

Disrupted Endothelial Tight Junction Protein Claudin-5 Recovered by Dexamethasone in a Mouse Model of Asthma

Background: The tight junction (TJ) protein, claudin 5 (CLN5) is a critical regulator of TJ. But its clinical signifi cance and therapeutic trial of the molecular mechanisms underlying CLN5 in bronchial asthma remain unresolved. To identify the role and reg- ulator of the endothelial tight junction protein CLN5 in a mouse model of asthma.

Methods: Using mice sensitized with ovalbumin (OVA) and challenged with OVA (OVA mice) as well as mice sensitized with saline and challenged with air (sham mice), we investigated potential links of CLN5 and bronchial asthma using RT-PCR, ELISA, immu- noblotting, and confocal imaging.

Results: In OVA mice, infl ammatory cytokine levels, airway infl ammation, and bron- chial hyperresponsiveness were increased than in sham mice. Lung CLN5 transcript and protein level were increased in OVA mice than in sham mice. Lung endothelial cells integrity was disrupted in OVA mice compared with sham mice. Dexamethasone treatment recovered disrupted CLDN5

Conclusions: These results indicate that CLN5 be implicated in the pathogenesis of bronchial asthma and represent a potential target for therapeutic intervention.

A Six-Year Study on the Changes in Pollen Counts and Sensitization Rates in South Korea

Background: The amount of airborne pollen is considered to be related to sensitiza- tion rates. The sensitization rates to tree pollen signifi cantly increased and those to grass and weed pollen decreased over the last 30 years in South Korea. We evaluated short term changes of sensitization rates and the correlation with pollen counts.

Methods: We assessed the number of pollen grains collected in Seoul. Tree pollen of the 15 species, weed pollen of the 8 species and grass were studied from January 1, 2007 to December 31, 2013. We retrospectively reviewed the results of 4,442 skin- prick tests (SPTs) to 33 common inhalant allergens conducted in the Severance Hospi- tal allergy clinics in Seoul from January 1, 2008 to December 31, 2013.

Results: The pollen counts of tree, especially pine and corylus, and grass signifi cantly decreased, as years go by. SPT results showed that the skin reactivity to grass sig- nifi cantly increased from 6.9% in 2008 to 14.0% in 2013. The sensitization rates to tree and weed pollen had no changes. In the details, the sensitization rates to Walnut, Popular, Bermuda, Timothy and Dandelion signifi cantly increased. There was signifi cant correlation between rate of change in pollen counts and sensitization rates to oak and hop J (Humulus japonicus). However, the others showed no correlation.

Conclusions: Decrease tendency of pollen counts was observed in Seoul, over the seven years. Some species showed signifi cant correlation between pollen counts and sensitization rates over the six years.

A Network Modelling: New Insights into Longitudinal Trend in Allergen Sensitization

Background: So far, longitudinal studies evaluating a trend in atopic sensitization have simply compared positive rates among allergens. Network modelling is capable of visualizing co-sensitization patterns from the view-point of a system. In this study, we aimed to search longitudinal trends in co-sensitization patterns among inhalant allergen using a network modelling.

Methods: We retrospectively reviewed results of skin prick tests performed in the Allergic Clinic at the Seoul National University Hospital, Seoul Korea, in 1983, 1990, 1997, 2005, and 2012. The skin test positivity was defi ned when the wheal size of allergen was greater than that of histamine. Then we visualized co-sensitization pat- terns among inhalant allergens in each year using qgraph package of R software.

Results: Positive rates of individual inhalant allergens showed wide variations over time. However, co-sensitization of house dust mite and tree pollen has increasingly occurred, which led to approximate two allergens closely in a theoretical plane of an allergen system.

Conclusions: Given that it is less likely that house dust mite and tree pollen have common allergenic determinants, an increasing tendency in co-sensitization of both allergens over time may refl ect environmental changes in Korea.
