Snake Venom synergized Cytotoxic Effect of Natural Killer Cells on NCI H358 Human Lung Cancer Cell

[Abstract]
Objectives : Iinvestigatedwhethersnakevenomcansynergisticallystrengthenthecytotoxic effectsofNK-92cells,andenhancetheinhibitionofthegrowthoflungcancercellsincluding NCI-H358throughtheinductionofdeathreceptordependentextrinsicapoptosis.
Methods : SnakevenomtoxininhibitedcellgrowthofNCI-H358Cellsandexertednoninfluence onNK-92cellviability.Moreover,whentheywereco-culturedwithNKcellsandconcomitantly treatedwith4㎍/㎖ ofsnakevenomtoxin,moreinfluencewasexertedontheinhibitionof growthofNCI-H358cellsthanBVorNKcellco-culturealone.
Results : TheexpressionofFas,TNFR2andDR3andinNCI-H358lungcancercellswassignif- icantlyincreasedbyco-cultureofNK-92cellsandtreatmentof4㎍/㎖ ofsnakevenomtoxin, comparedtoco-cultureofNK-92cellsalone.Coincidentally,Bax,caspase-3andcaspase- 8-expressionsofpro-apoptoticproteinsintheextrinsicapoptosispathway,demonstrated significantincrease.However,inanti-apoptoticNF-κBactivities,activityofthesignalmolecule wassignificantlydecreasedbyco-cultureofNK-92cellsandtreatmentof4㎍/㎖ ofsnake venomtoxin,comparedtoco-cultureofNK-92cellsorsnakevenomtoxintreatedbyNCI- H358alone.Meanwhile,intermsofNOgeneration,thereisasignificantincrease,inco-culture ofNK-92cellswithNCI-H358cellsaswellastheco-cultureofNK-92cellsandconcomitant treatmentof4㎍/㎖ ofsnakevenomtoxin.However,nosynergisticincreaseofNOgeneration wasshowninco-cultureofNK-92cellsandtreatmentof4㎍/㎖ ofsnakevenomtoxin,com- paredtoco-cultureofNK-92cellswithNCI-H358cells.
Conclusion : Consequently,thisdataprovidesthatsnakevenomtoxincouldbeusefulcandidate compoundstosuppresslungcancergrowthalongwiththecytotoxiceffectofNK-92cells throughextrinsicapoptosis.
