that in vitro culture of pluripotent stem cells on laminin- (Domogatskaya et al., 2008) or vitronectin- (Kaini et al., 2016) coated culture plates can stimulate the maintenance of self-renewal. In contrast, we propose that integrins α 6 β 1 and α V β 1 may play more pivotal roles in initiating early stage differentiation of undifferentiated hybrid B6D2F1 mouse ESCs than in maintaining self-renewal, supported by the fact that there are many problems in differentiation using laminin signaling-free ICM during embryogenesis (Li et al., 2002) and mouse ESCs cultured on laminin or vitronectin easily lose their pluripotency (Heydarkhan- Hagvall et al., 2012, Hayashi et al., 2007). Those contradict statement for laminin and vitronectin in maintaining pluripotency or initiating early differentiation could be occurred by different stage of ESCs/blastocyst or by their diverse genetic background. Further studies on the distribution of ECM proteins in blastocysts and their cell to ECM communications are required to confirm this.

The aging skin microenvironment dictates stem cell behavior

Aged HFSCs Can Be Rejuvenated by Neonatal Dermis during In Vivo Transplantation. Our culture studies suggested that growth- promoting microenvironments might override intrinsic age-related deficiencies in at least some HFSCs within the population. To evaluate the fitness of the aging HFSC population as a whole, and in a physiological context, we turned to HFSC engraftment studies. Previous studies on young skin showed that following culture, HFSCs can regenerate epidermis and HFs when they are combined with freshly isolated neonatal dermal cells and then engrafted onto the back skins of immune-deficient recipient mice (39, 64, 66–69). We were concerned that culture might arti- ficially select for HFSCs most able to withstand culture stress, hence confounding age-related differences. To circumvent this caveat, we omitted the culture step, and performed the engraft- ments with FACS-purified HFSCs freshly isolated from aged and young skins (Fig. 7A; see Materials and Methods for further details).

However, it was unclear why steroid production decreased during the early stage of follicle differentiation in vitro. Such aberrant secretion may result from differences between the in vivo and in vitro environments, or an error may have been made in hormone measurement, as P4 levels were lower than those reported in other studies. Our standard laboratory-operation protocol was employed for sample collection, storage, and measurement of the hormones, and the experimental design and management was strictly executed to avoid data errors between treatments. Accordingly, the presence of fewer granulosa cells in primary than in secondary follicles may have resulted in the reduced hormone production observed. The lower P4 concentrations could also be due to the use of supernatants for the analysis or to different operating protocols. In this study, we used conditioned media to non-invasively analyze hormone secretion. The level of P4 in conditioned medium was less than the minimal level of detection. In our preliminary experiment, the minimal concentration of P4 was similar to that of E2.

In addition to biomaterials for 3D culture, the development of potent enhancers of stem cell func- tions, including small molecules, biologics, or genes, may greatly facilitate stem cell engineering or priming pro- cesses prior to clinical use. Although we showed that this 3D priming strategy of human SGSCs is attractive for fu- ture clinical applications, further studies are necessary to determine the optimal timing and delivery of SGSCs 3D , as well as to identify the key trophic factors released from SGSCs 3D , and the related molecular pathway mediating the radioprotective effects for future clinical applications.

Given the fact that donor-derived stromal cells do not engraft following conventional HSCT due to the limited number of stromal cells present in a typical allograft even having a competitive advantage over the damaged recipient's stroma, donor stromal cell engraftment could be achieved by transplanting a large number of MSCs that had been expanded in culture 8 . Previous studies in mice and fetal sheep have demonstrated the capacity of marrow stromal cells to engraft if administered in sufficient quantity, providing additional support for this premise 9,10 . Recent researches have demonstrated that cotransplantation of human MSCs along with human HSCs enhance human hematopoietic engraftment in animal models 11~14 and in human 15,16 . Moreover, MSCs possess in vitro immunosuppressive properties that appear not to be the major histocompatibility complex restricted 17,18 . In these respects, cotransplan- tation of allogeneic MSCs with allogeneic HSCs can provide some beneficial effects such as the enhancement of engraftment and suppression of graft-versus-host activity 19 . However, a possible negative influence of MSCs' immunosuppressive properties on graft-versus-leukemia effect in allograft setting has not been mentioned. In vitro stromal cell dependent growth of leukemic cells also suggests the possibility of beneficial effect on leukemia survival following MSCs transplantation.

iii expression of inserted lentiviral transgenes was controlled by DNA methylation. Neuronal progenitor cells were derived from pig embryonic germ cells. Similar with other species, neuronal progenitor cells were successfully induced by treatment of retinoic acid and these cells expressed neuronal markers such as PAX6, NESTIN and SOX1 . Taken together, I found that, as a non-permissive species, pig PSCs are maintained by mainly FGF signaling, and additional signaling molecules such as ACTIVIN and WNT are required for supporting pluripotency. And pig ESCs could be derived using chemically defined media supplementing FGF2, ACTIVIN A and WNT. This study will not only provide basic understanding for mechanism of maintaining pluripotency but also apply stem cell engineering for regenerative medicine. Accordingly, studies on pig PSCs will pave the way for human cell therapy and shed new light on researches of PSCs.

Effect of EDTA on canine parthenote development during in vitro culture

Abstract Somatic cell nuclear transfer (SCNT) is a useful biotechnological tool for animal cloning. Until now, SCNT has been inefficient, especially in dog. It is believed that an embryo developmental block in SCNT embryos is cause of low production efficiency. However, no studies have been performed on canines for embryo developmental block. In this study, we attempted to evaluate the beneficial role of EDTA in canine parthenogenic (PA) embryos development to overcome embryo developmental block. The PA embryos were divided into 0.01 mM EDTA treated and non-treated groups. Embryo developmental efficiency was measured by activating chemically parthenote. After EDTA induction, PA embryos were evaluated for embryonic development, Reactive Oxygen Species (ROS) activity, mitochondrial integrity, ATP production and genomic activation. The EDTA treated PA embryos showed significantly higher survival rate and improved cavity formation compared to non-treated. Furthermore, cytoplasmic ROS level was mitigated and mitochondrial membrane potential was found significantly higher in EDTA treated group followed by higher ATP production. Moreover, major embryonic genomic activation specific markers/factors were also elevated in EDTA treated group. Conclusively, we elucidated that EDTA showed substantially positive effect to overcome embryo developmental block in canine.

In addition, the transplantation of BMCs, HSCs and MSCs is considered safe and has been widely tested in clinical trials in individuals with liver diseases with encouraging results. However, the development of cell therapy for cirrhosis requires larger clinical studies to obtain meaningful insights into the safety and clinical efficacy of cell infusion [81,82] . This is especially true in the case of MSC transplantation, for which reports on the efficacy of MSCs are controversial and are dependent on the research group; however, we expect that issues concerning the efficacy of these cells will be resolved in the near future by a large multicenter randomized clinical trial that is currently being conducted in Korea. Moreover, multicenter international clinical studies on the safety and efficacy of MSC treatments for cirrhosis can help clinicians reach a consensus regarding the treatment of liver fibrosis, which will ultimately improve the prognosis of patients.

Plasma gel (PG) is a biologic gel derived from blood; the plasma is easily obtained by blood sampling and cen- trifuge. PG is autologous material with notable advantages in clinical applications over other scaffolds; less risks of infection or rejection reaction and no necessary of separate FDA approval. In terms of fabrication methods, studies have been published on mixing PG with CaCl 2 , fibrin, other gels including hydrogel, and hyaluronic acid as well as heating it 9–13 ; among these methods, heating has the advantages of shorter production time and lower cost. For cosmetic augmentation and vocal fold paralysis treatment, plasma gel fabricated by heating has been used, and its safety was demonstrated 12,14 . Plasma was also reported to provide excellent conditions for cell culture and differentiation because of the abundance of growth factors and ECM 10,15 .

Therefore, in this study, in order to find out how the stem cell culture medium is heated or preserved at high temperature, the effect of various effects of stem cells on the various effects of the stem cells was investigated. As a result of the experiment, the wound healing assay confirmed that the cell migration increased after 6 hours, and after 24 hours, it was confirmed that the cell mobility was increased and cell division was promoted, thereby being concentrated. As a result of investigating the amount of transdermal water loss by preparing a cosmetic product containing stem cell culture solution, it was confirmed that the culture solution addition group showed an improvement rate of 31% compared to the non-added group, thereby helping in skin wound recovery. As a result of this, it is considered that this point should be considered when the stem cell culture medium is used as an active ingredient in cosmetics in the future.

Mesenchymal stem cells (MSCs) are emerging as an attractive option for osteoarthritis (OA) of the knee joint, due to their marked disease-modifying ability and chondrogenic potential. MSCs can be isolated from various organ tissues, such as bone marrow, adipose tissue, synovium, um- bilical cord blood, and articular cartilage with similar phenotypic characteristics but different proliferation and differentiation potentials. They can be differentiated into a variety of connective tissues such as bone, adipose tissue, cartilage, intervertebral discs, ligaments, and muscles. Al- though several studies have reported on the clinical efficacy of MSCs in knee OA, the results lack consistency. Furthermore, there is no consensus regarding the proper cell dosage and application method to achieve the optimal effect of stem cells. Therefore, the purpose of this study is to re- view the characteristics of various type of stem cells in knee OA, especially MSCs. Abstract This paper is intended to apply Mugunghwa, which is the official flower that symbolizes the country, to fashion cultural products to promote the identity and enhance the cultural value of South Korea. The purpose of this study is to increase the understanding and use of tradition. As for study methods, first, the range of uses for Mugunghwa were examined from diverse angles, second, images associated with modern senses that reflect trends were developed to create more value. Third, the role of a cultural ambassador was proposed by adding the image of Mugunghwa to fashion accessories and miscellaneous goods applicable to real life. This was carried out through theoretical and empirical studies, such as literature reviews and case study analysis, and as a result, 13 textile designs and 26 fashion cultural products were developed. It is hoped that the foregoing will enable the pursuit of new values of the image symbolizing the country and the forming a bond, and it is expected that the foregoing can provide opportunities to develop diverse national symbols, thereby enhancing the national image.
